蛋白结构:
该蛋白在C端带有多组氨酸标签。补充C5的成熟形式是由蛋白水解裂解的α和β链组成的二硫键连接的异二聚体。每个α和β链的计算MW分别为73.9kDa(β链)和114.4kDa(α链)。在还原(R)条件下(SDS-PAGE),蛋白质以67kDa(β链)和106-116kDa(α链)迁移,在非还原(NR)条件下,由于糖基化,蛋白质以130-150kDa迁移。

分子背景:
C5过敏毒素来源于补体C5的蛋白水解降解,是局部炎症过程的媒介。C5前体通过去除4个碱性残基进行处理,形成由二硫键连接的两条链,β和α。C5转化酶通过裂解α链激活C5,释放C5a过敏毒素并产生C5b(β链+α’链)。通过C5转换激活C5启动晚期补体成分C5-C9自发组装成膜攻击复合物。C5b具有C6的瞬时结合位点。C5b-C6复合物是裂解复合物组装的基础。C5a过敏毒素与C5AR1和蜱补体抑制剂相互作用。C5a也是一种有效的趋化因子,它刺激多形核白细胞的运动,并引导其向炎症部位迁移。
